Background
This product is an unconjugated, non-therapeutic recombinant analog of the anti-interferon-gamma antibody fontolizumab, engineered as a human IgG1/kappa immunoglobulin that binds the soluble cytokine IFN-gamma. It is supplied for research use only and is not the clinical drug; it is not intended for human or veterinary use. Built around the IFN-gamma target, it functions as a neutralizing (cytokine-blocking) antibody that engages IFN-gamma and prevents it from signaling through its receptor. Because it is offered as a research-grade reagent, it is well suited to work that requires a defined, reproducible anti-IFN-gamma binder: isotype-matched positive controls, IFN-gamma neutralization assays, target-engagement and epitope studies, standardization of cytokine bioassays, and preclinical characterization workflows. It is available at low endotoxin levels (research grade typically under 1 EU/mg, with ultra-low-endotoxin options for sensitive cell-based and in-vivo standard applications) and can be produced in bulk milligram-to-gram quantities to support scale-up, assay development, and lot-bridging. This makes it a practical, cost-effective substitute for the originator molecule in discovery and method-development settings.
Interferon-gamma (IFN-gamma), encoded by IFNG (UniProt P01579), is the sole type II interferon. It is a homodimeric cytokine produced mainly by activated T helper 1 (Th1) cells, CD8-positive cytotoxic T cells, natural killer cells, and NKT cells. IFN-gamma signals through a receptor complex of IFNGR1 and IFNGR2, activating the JAK1/JAK2-STAT1 pathway and driving transcription of interferon-stimulated genes. Its biological effects include activation of macrophages and enhanced microbial killing, upregulation of MHC class I and class II antigen presentation, promotion of Th1 differentiation, and modulation of both innate and adaptive immunity. IFN-gamma is central to host defense against intracellular pathogens and to tumor immunosurveillance, but dysregulated IFN-gamma also contributes to chronic inflammatory and autoimmune conditions. This dual protective/pathogenic role makes IFN-gamma a long-standing target for both neutralization and immunomodulation research.
