Background
ICH1043 is a purified rat anti-mouse CD8 monoclonal antibody, clone YTS-169, of the rat IgG2b isotype. YTS-169 recognises mouse CD8 (CD8 alpha) and is a widely used reagent for the in vivo depletion of CD8-positive cytotoxic T lymphocytes, making it well suited to functional studies that dissect the contribution of CD8 T cells to anti-tumour immunity, infection, autoimmunity and transplant responses. The clone is also validated for flow cytometry and IHC on frozen sections, allowing depletion efficiency to be confirmed on the same material. This preparation is manufactured to support in vivo work: it is supplied at low endotoxin levels (research-grade less than 1 EU/mg; ultra-low options less than 0.5 EU/mg) to minimise confounding innate immune activation, is available in bulk milligram-to-gram quantities suitable for repeat-dosing cohort studies, and is provided for research use only. Because researchers typically search by clone, note that YTS-169 is frequently paired with the non-depleting or complementary anti-CD4 clones in T-cell subset depletion experiments.
CD8 is a cell-surface glycoprotein that serves as the co-receptor for the T-cell receptor (TCR) on major histocompatibility complex (MHC) class I-restricted T lymphocytes. It is expressed predominantly as an alpha-beta heterodimer on cytotoxic T cells, with an alpha-alpha homodimer form on some dendritic and other cells. During antigen recognition, the CD8 alpha chain binds the conserved alpha-3 domain of MHC class I, stabilising the TCR-peptide-MHC interaction and recruiting the tyrosine kinase Lck to the intracellular signalling complex, thereby lowering the threshold for T-cell activation. CD8-positive cytotoxic T lymphocytes are central effectors of adaptive immunity, killing virally infected, intracellular-pathogen-infected and tumour cells through perforin/granzyme release and Fas-mediated pathways, and are a principal target of many cancer immunotherapies. The clone here targets the CD8 alpha chain.
