Background
ICH5338 is a research-grade biosimilar of alomfilimab, an unconjugated, non-therapeutic analog of the anti-ICOS antibody originally developed as KY1044 (later SAR445256). It is a human IgG1 kappa antibody built around the inducible T-cell co-stimulator (ICOS, CD278) and is intended for research use only; it is not the clinical drug and is not for human or diagnostic use. The originator was designed for a dual mechanism in oncology: agonism of ICOS on effector T cells alongside antibody-dependent cellular cytotoxicity (ADCC)-mediated depletion of ICOS-high regulatory T cells, exploiting the differential expression of ICOS across T-cell subsets to reshape the tumor immune microenvironment. This biosimilar reproduces the antigen-binding specificity of the originator so that laboratories can study ICOS engagement, co-stimulatory signaling, and Fc-dependent effector functions in controlled settings. It is supplied at research grade with low endotoxin, in a format suitable for binding, blocking, neutralization, ADCC, and reporter assays, and as a reference or benchmarking reagent. Because ICOS is a human target, the antibody is primarily suited to in-vitro and functional characterization rather than mouse in-vivo dosing. Specifications should be confirmed against the current datasheet. ICOS (inducible T-cell co-stimulator, CD278) is a CD28-family co-stimulatory receptor expressed on activated T cells. It is induced following T-cell activation and binds its ligand ICOS-L (B7-H2, CD275) on antigen-presenting cells, delivering a co-stimulatory signal that supports T-cell proliferation, cytokine production, and effector differentiation, including follicular helper T-cell responses and germinal center function. ICOS is expressed at high levels on immunosuppressive regulatory T cells and at lower levels on effector T cells, and this differential expression underpins interest in ICOS as an immuno-oncology target. Agonism of ICOS can enhance effector T-cell activity, while antibodies with active Fc regions can preferentially deplete ICOS-high regulatory T cells through ADCC. This combination of co-stimulatory signaling and effector-cell modulation makes ICOS a focus for cancer immunotherapy research and for studies of T-cell tolerance and activation.
