Background
This is a research-grade, unconjugated recombinant analog of uliledlimab, a human IgG1 (kappa) anti-CD73 (NT5E) antibody built around the well-characterized CD73 target. It is a non-therapeutic biosimilar supplied for research use only and is not the clinical product; it is not intended for human or veterinary use. The molecule carries an N297A substitution in the Fc region, which abolishes N-linked glycosylation at that site and effectively silences Fc gamma receptor engagement and antibody-dependent effector function, making it useful as an effector-minimized reagent for isolating target-binding and enzyme-blocking effects from Fc-driven activity. Typical applications include CD73 binding and neutralization assays, comparison against other anti-CD73 tool antibodies, use as a reference or benchmark control in adenosine-pathway immunology, and antibody-engineering and antibody-drug-conjugate development workflows. It is offered at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg available), supporting sensitive cell-based work, and can be provided in bulk milligram-to-gram quantities for larger screening or in-vitro standardization programs.
NT5E encodes ecto-5'-nucleotidase (CD73), a GPI-anchored cell-surface glycoprotein that functions as a homodimer. It is the rate-limiting enzyme that hydrolyzes extracellular AMP to adenosine, working downstream of CD39 (which converts ATP/ADP to AMP). The adenosine generated by CD73 acts through A2A and A2B adenosine receptors and is broadly immunosuppressive: it dampens effector T-cell proliferation and cytokine output, supports regulatory T-cell function, and modulates myeloid, endothelial, and epithelial cells. CD73 is widely expressed across tissues and on lymphocyte subsets, macrophages, dendritic cells, and endothelium, and is frequently upregulated in the tumor microenvironment, where the CD39-CD73-adenosine axis contributes to immune escape. Loss-of-function NT5E mutations cause arterial calcification (ACDC), underscoring the enzyme's role beyond immunity in vascular biology.
