Background
This product is an unconjugated, non-therapeutic recombinant analog of dresbuxelimab (originally AK-119), a human IgG1-kappa monoclonal antibody directed against CD73 (NT5E; UniProt P21589). It is manufactured for research use only and is not the clinical drug, nor is it intended for human or veterinary use. Built around the CD73 target, it reproduces the binding specificity of the originator so investigators can study the CD73/adenosine axis in controlled settings. Typical applications include use as a positive/benchmark control in binding and enzyme-inhibition assays, as a reference reagent in immunoassay development and epitope-competition studies, and as a tool for antibody-engineering and ADC/ADCC-development workflows. The antibody is offered at low-endotoxin research grade (<1 EU/mg), with an ultra-low-endotoxin option (<0.5 EU/mg) suited to sensitive cell-based and preclinical work, and is available in bulk milligram-to-gram quantities for scaled or longitudinal studies. Supplying a consistent, well-characterised biosimilar analog lets labs compare data against a stable reference lot rather than variable in-house reagents. CD73, encoded by NT5E, is a glycosylphosphatidylinositol (GPI)-anchored ecto-5'-nucleotidase that acts as the rate-limiting enzyme in extracellular adenosine generation. It hydrolyses extracellular AMP to adenosine and inorganic phosphate, working downstream of CD39 (ENTPD1), which converts ATP/ADP to AMP. The resulting adenosine engages P1 receptors (notably A2A and A2B) on immune cells, producing broadly immunosuppressive effects: dampened T-cell and NK-cell activation, and promotion of regulatory and myeloid-suppressor phenotypes. CD73 is expressed on many stromal, endothelial, and epithelial cells, and is frequently upregulated in the tumor microenvironment, where the adenosinergic pathway contributes to immune evasion, angiogenesis, and metastasis. Beyond oncology, CD73-derived adenosine regulates vascular tone, barrier function, and inflammation. These roles make CD73 a prominent target for research into tumor immunology and adenosine-pathway modulation.
