Background
This product is an unconjugated, non-therapeutic recombinant analog of simtuzumab, supplied for research use only. Simtuzumab is a humanized IgG4 monoclonal antibody directed against lysyl oxidase-like 2 (LOXL2), the enzyme it was originally developed to inhibit in fibrotic and oncology settings. The research-grade analog reproduces the antigen-binding specificity of the originator against human LOXL2 (UniProt Q9Y4K0) and is intended as a tool reagent rather than a clinical product. It is useful as a defined anti-LOXL2 binder for in-vitro and preclinical work: as a positive-control or reference antibody in binding and enzyme-inhibition assays, for probing LOXL2-dependent collagen and elastin cross-linking, as a benchmark in biosimilar or comparability studies, and as a starting scaffold for ADC or engineering development. The human IgG4 backbone gives minimal effector function, which suits neutralization-focused experiments. It is offered at research grade with low endotoxin, and bulk milligram-to-gram quantities are available for larger studies. It is not the clinical drug and is not for human or veterinary use.
Lysyl oxidase-like 2 (LOXL2) is a secreted, copper- and lysyl-tyrosylquinone-dependent amine oxidase belonging to the lysyl oxidase (LOX/LOXL) family. Its canonical catalytic role is oxidative deamination of peptidyl lysine and hydroxylysine residues in collagen and elastin, initiating covalent cross-link formation that stabilizes and stiffens the extracellular matrix. Through this activity LOXL2 contributes to matrix remodeling in tissue fibrosis (hepatic, pulmonary, renal, cardiac) and to the desmoplastic, stiffened stroma of many solid tumors. LOXL2 also has intracellular and non-enzymatic functions, including regulation of epithelial-to-mesenchymal transition, in part by modulating transcription factors such as SNAIL, and roles in angiogenesis and metastatic niche formation. Its expression is frequently elevated in fibrotic disease and in carcinomas, where high levels associate with progression, making it a longstanding target for anti-fibrotic and anti-tumor strategies.

Specificity

Purification Method
This monoclonal antibody was purified using multi-step affinity chromatography methods such as Protein A or G depending on the species and isotype.

This biosimilar is stable when stored at 2-8°C. For longer term storage (> 3 months) it is recommended to store this antibody at -20°C or -70°C.
