Background
ICH5084 is a research-grade, unconjugated recombinant analog of Rolinsatamab, a monoclonal antibody directed against the human prolactin receptor (PRLR). It is supplied as a human IgG1-kappa protein carrying an engineered S239C cysteine substitution in the Fc region, a common site used to enable site-specific payload conjugation, making this molecule a useful naked-antibody scaffold for antibody-drug conjugate (ADC) development workflows. This product is a non-therapeutic biosimilar intended strictly for research use only (RUO); it is not the clinical drug and is not for human or veterinary use. It is offered unconjugated so investigators can characterize target binding and downstream biology, or perform their own conjugation chemistry. Because it is manufactured to research standards with low endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg) and is available in bulk milligram-to-gram quantities, it suits reproducible in-vitro assays, binding and blocking studies, isotype-matched control comparisons, and preclinical model development. PRLR (prolactin receptor; UniProt P16471) is a single-pass type I transmembrane receptor of the class I cytokine receptor superfamily, encoded on human chromosome 5p13-14. Its principal ligand is the anterior pituitary hormone prolactin, though it also binds growth hormone and placental lactogen. Ligand engagement drives receptor dimerization and activation of receptor-associated Janus kinase 2 (JAK2), initiating JAK-STAT signaling with STAT5 as the dominant transducer, alongside MAPK and Src-family kinase pathways. PRLR signaling governs mammary lobuloalveolar development, lactation, reproduction, and cell proliferation and differentiation. Because prolactin signaling can promote survival and growth in breast and other epithelial tissues, PRLR is studied as a candidate target in PRLR-positive cancers, which motivates interest in PRLR-directed antibodies and antibody-drug conjugates.
