Background
The 176F (low-affinity) allotype of human CD16a (FcγRIIIa) is a major activating Fc gamma receptor on NK cells and macrophages that drives antibody-dependent cellular cytotoxicity, and allelic variation at this position strongly influences therapeutic-antibody potency. This recombinant CD16a (176F) ectodomain, expressed in HEK 293 cells, is validated for SPR, BLI and ELISA to support Fc binding kinetics, ADCC characterization and Fc-variant ranking. It is a research-use-only protein supplied in an in vivo grade, ultra-low-endotoxin format and produced at scalable milligram (mg) to gram (g) scale, giving high-throughput and bulk-screening laboratories a dependable allotype-defined CD16a reagent.

Specificity
The sequence of the extracellular domain of human CD16a (Gly 17-Gln 208) was fused with a C-terminal tag consisting of the AVI tag, TEV protease recognition sequence and a 10-His tag.
Formulation
Lyophilized from sterile PBS, pH 7.4. No preservatives or cryoprotectants have been added. To obtain a final concentration of 1 mg/ml reconstitute 0.25 mg vials with 0.25 ml water and 1.0 mg vials with 1.0 ml water.
