Background
This is a research-grade, unconjugated recombinant analog of glembatumumab, a monoclonal antibody directed against human GPNMB (glycoprotein non-metastatic melanoma protein B, also known as osteoactivin; UniProt Q14956). It is a non-therapeutic, research-use-only reagent and is not the clinical drug; it is not intended for human or veterinary use. The originator antibody was advanced clinically as an antibody-drug conjugate (glembatumumab vedotin) exploiting GPNMB overexpression on tumor cells and its capacity for internalization. This unconjugated analog reproduces the antigen-binding specificity for GPNMB without any cytotoxic payload, making it useful as a target-binding tool, a positive/isotype-matched control, a naked-antibody comparator in ADC development, and a reagent for characterizing GPNMB expression and antibody-antigen interactions in vitro. It is supplied as a full-length human IgG2-kappa immunoglobulin. Low-endotoxin preparations (research grade <1 EU/mg; ultra-low <0.5 EU/mg) and bulk milligram-to-gram quantities support assay development, binding and neutralization studies, and preclinical model work where a consistent, well-characterized anti-GPNMB reagent is needed.
GPNMB is a type I transmembrane glycoprotein whose extracellular domain can be shed by proteases (e.g., ADAM10) to release a soluble ectodomain. It is expressed in melanocytes, osteoclasts, dendritic cells, macrophages, and various epithelial tissues, and is markedly overexpressed in several cancers including melanoma, triple-negative breast cancer, and gliomas, where high expression associates with invasion and poor prognosis. Functionally, GPNMB promotes cell adhesion, migration, and tissue regeneration, and modulates immune responses, acting as a negative regulator of T-cell activation and dampening inflammation in macrophages. It also participates in osteoblast differentiation and bone matrix mineralization. Because it is surface-exposed, internalizes upon antibody binding, and is enriched on tumor cells relative to normal tissue, GPNMB has been pursued as an antibody-drug-conjugate target. Its dual roles in tumor biology and immune modulation make it a subject of ongoing research.
