Background
This is an unconjugated, non-therapeutic recombinant analog of etrolizumab, supplied strictly for research use only. It is not the clinical drug and is not intended for human or veterinary use. Etrolizumab is an anti-integrin antibody engineered around the β7 integrin subunit; the originator was investigated in inflammatory bowel disease. This research-grade analog reproduces the binding specificity of the originator (a human IgG1 format) so investigators can study integrin-mediated adhesion and leukocyte trafficking in controlled settings. Typical applications include use as a positive/reference control in binding and neutralisation assays, as a benchmarking reagent alongside other anti-integrin tools, in ADCC/effector-function characterisation, and as a starting point for assay development or biosimilarity comparisons. It is offered at research-grade low endotoxin (<1 EU/mg), with an ultra-low endotoxin option (<0.5 EU/mg) suitable for sensitive cell-based work, and in bulk milligram-to-gram quantities to support scale-up, screening campaigns, and preclinical model work. Formulation is designed to be flexible for downstream reformatting.
Integrins are heterodimeric adhesion receptors built from paired alpha and beta subunits. ITGB7 (integrin beta-7, P26010) partners with two distinct alpha chains: ITGA4 (integrin alpha-4, P13612) to form alpha4beta7, and integrin alphaE (ITGAE) to form alphaEbeta7. Alpha4beta7 is expressed on gut-homing lymphocytes and binds MAdCAM-1 on intestinal vascular endothelium, directing lymphocyte trafficking into gut-associated lymphoid tissue; it also binds VCAM-1 and fibronectin. AlphaEbeta7 is expressed on intraepithelial and tissue-resident lymphocytes and binds E-cadherin, promoting retention within epithelial layers. The shared beta-7 subunit therefore governs both mucosal recruitment and epithelial retention of leukocytes, making it a central node in gut mucosal immunity. Antibodies directed at the beta-7 subunit can simultaneously interfere with both alpha4beta7 and alphaEbeta7 function, distinguishing them from reagents that target the alpha4 chain alone.
