Background
This is a research-grade, unconjugated recombinant analog of enibarcimab (formerly adrecizumab), an antibody built around human adrenomedullin (ADM). It is a non-therapeutic biosimilar intended for research use only (RUO) and is not the clinical drug; it is not for human or veterinary use. The molecule is a full-length human IgG1-kappa (carrying the E356D/M358L allotype substitutions in the Fc) that recognizes the ADM peptide, making it a useful tool reagent for target-engagement studies, standard curves, positive/isotype-matched controls, assay development, and in-vitro and preclinical characterization of ADM biology. Because it reproduces the antigen-binding behavior of the originator without therapeutic intent, researchers can use it to benchmark binding, screen assay formats, and support ADCC/ADC-development-style workflows independent of a clinical supply. It is supplied unconjugated to preserve flexibility for downstream labeling or immobilization. Adrenomedullin (ADM; gene ADM, UniProt P35318) is a 52-amino-acid vasoactive peptide derived from preproadrenomedullin, which is also processed to proadrenomedullin N-terminal 20 peptide (PAMP). ADM belongs to the calcitonin/CGRP peptide family and signals through the calcitonin receptor-like receptor (CALCRL) in complex with receptor activity-modifying proteins RAMP2 and RAMP3, driving cAMP production. It is produced by vascular endothelial and smooth muscle cells and many other tissues, and is a key regulator of vascular tone and, importantly, endothelial barrier integrity. ADM promotes vasodilation, counteracts vascular leakage, and has roles in blood pressure control, cardiovascular homeostasis, and the response to inflammatory and septic insults. Circulating bioactive ADM is a recognized biomarker of endothelial dysfunction; both excess and dysregulated ADM signaling are implicated in septic shock, heart failure, and other critical illness.
