Background
Carrying the higher-affinity 176V polymorphism, this allotype of human CD16a (FcγRIIIa) is central to NK-cell and macrophage antibody-dependent cellular cytotoxicity and is widely studied because the V/F polymorphism modulates clinical antibody responses. The recombinant CD16a (176V) ectodomain is expressed in HEK 293 cells and validated for SPR, BLI and ELISA, enabling precise Fc binding kinetics and side-by-side comparison of Fc engineering strategies. Offered for research use only as an in vivo grade, ultra-low-endotoxin preparation, it is manufactured in scalable milligram (mg) to gram (g) quantities so that bulk and high-throughput laboratories can profile allotype-specific Fc interactions reproducibly.

Specificity
The sequence of the extracellular domain of human CD16a (Gly 17-Gln 208) was fused with a C-terminal tag consisting of the AVI tag, TEV protease recognition sequence and a 10-His tag.
Formulation
Lyophilized from sterile PBS, pH 7.4. No preservatives or cryoprotectants have been added. To obtain a final concentration of 1 mg/ml reconstitute 0.25 mg vials with 0.25 ml water and 1.0 mg vials with 1.0 ml water.
