Background
This is clone TA99 (also written TA-99), a mouse monoclonal of the IgG2a, kappa isotype raised against TYRP1/TRP1 (gp75), the product of the mouse brown (b) locus. Because it is a mouse IgG2a, the antibody engages activating Fc-gamma receptors and complement effectively, making it well suited to Fc-dependent functional and in-vivo antitumour studies in immunocompetent mice as well as to biochemical and imaging applications. It is broadly validated across ELISA, immunoprecipitation, immunocytochemistry, immunofluorescence, immunohistochemistry, radioimmunoassay and functional/in-vivo assays, and per the product record reacts with mouse and human TYRP1 (UniProt P07147). Supplied as a low-endotoxin, bulk-scale (milligram to gram) research-grade preparation for research use only, it is intended for laboratories running antibody-dependent effector, tumour-immunology, melanocyte-biology and pigmentation experiments where carryover endotoxin and lot-to-lot consistency matter. Low endotoxin and bulk formatting make it appropriate for repeat-dosing in-vivo work and larger in-vitro campaigns without reformulation. TYRP1 (tyrosinase-related protein 1), also called TRP1 or gp75, is a melanosomal type-I membrane glycoprotein and one of the most abundant melanocyte proteins. It belongs to the tyrosinase family and contributes to the eumelanin synthesis pathway, where it participates in the oxidation of DHICA (5,6-dihydroxyindole-2-carboxylic acid) and stabilises and modulates the activity of tyrosinase within the melanosome; loss-of-function alleles produce brown rather than black coat colour in mice and a form of oculocutaneous albinism in humans. Because it is highly and selectively expressed by melanocytes and by most melanomas, and because part of the molecule is accessible at the cell surface, TYRP1/gp75 is a classic melanoma differentiation antigen and a target for antibody-mediated recognition. Its restricted, lineage-specific expression underlies its use as a melanocytic marker in histology and as an immunotherapy target antigen.
