Background
This product is an unconjugated, non-therapeutic recombinant analog of orilanolimab, an anti-FcRn (neonatal Fc receptor) monoclonal antibody, supplied strictly for research use only. It is not the clinical drug and is not intended for human or veterinary use. Built as a human IgG4-kappa molecule directed against the FCGRT-encoded alpha chain of FcRn, it is intended to reproduce the target-binding behaviour of the originator for laboratory investigation of FcRn biology and antibody-recycling pathways. Typical research applications include use as a binding and blocking reference standard, a positive control in FcRn-interaction and IgG-homeostasis assays, a comparator in biosimilar or developability workflows, and a tool for probing FcRn-mediated IgG and albumin transport in vitro. The material is offered at low-endotoxin research grade (<1 EU/mg), with ultra-low-endotoxin (<0.5 EU/mg) and in-vivo-standard preparations available, and can be produced in bulk milligram-to-gram quantities to support screening, formulation, and preclinical mechanistic studies. It carries no fluorophore, enzyme, or other conjugate. FCGRT encodes the alpha (heavy) chain of the neonatal Fc receptor (FcRn), an MHC class I-like glycoprotein that associates non-covalently with beta-2-microglobulin. FcRn is the central regulator of immunoglobulin G and serum albumin homeostasis. In a pH-dependent manner, it binds the Fc region of IgG and albumin within acidified endosomes (around pH 6), diverting them away from lysosomal degradation and recycling them back to the cell surface, where near-neutral pH triggers their release into circulation. This salvage pathway gives IgG and albumin their unusually long serum half-lives. FcRn also mediates transcytosis across epithelial and endothelial barriers, including maternal-fetal IgG transfer across the placenta, and contributes to antigen presentation. Blocking FcRn accelerates catabolism of circulating IgG, including pathogenic autoantibodies, which is the therapeutic rationale behind anti-FcRn antibodies in autoimmune disease.
