Background
This product is an unconjugated, non-therapeutic recombinant analog of gevokizumab, supplied for research use only. Gevokizumab (originally XOMA 052) is a humanized IgG1 monoclonal antibody directed against human interleukin-1 beta (IL-1B), notable for its allosteric mechanism: rather than fully blocking IL-1B from contacting its receptor, it modulates the cytokine's binding kinetics to the IL-1 receptor type 1 (IL-1R1)/IL-1RAcP signaling complex. This research-grade analog reproduces the target specificity of the originator and is intended as a laboratory reagent, not as a clinical or veterinary drug. It is useful as a defined anti-IL-1B tool for neutralization and binding assays, as a positive/isotype-matched control in IL-1 pathway experiments, for benchmarking against other IL-1B binders, and for antibody-engineering and ADC/bispecific development workflows. It is offered in low-endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg) formats and in bulk milligram-to-gram quantities to support in-vitro assay development and preclinical screening. It is not the clinical product and is not intended for human use.
Interleukin-1 beta (IL-1B, UniProt P01584) is a prototypic pro-inflammatory cytokine of the IL-1 family. It is synthesized as an inactive 31 kDa precursor (pro-IL-1B) that is cleaved by caspase-1 within the inflammasome to yield the mature ~17 kDa active cytokine, which is then released from monocytes, macrophages, and other cells. Mature IL-1B signals through the type 1 IL-1 receptor (IL-1R1) together with the accessory protein IL-1RAcP, recruiting MyD88 and driving NF-kB and MAPK activation. This induces downstream mediators including IL-6, COX-2/prostaglandins, and further IL-1, amplifying local and systemic inflammation, fever, and leukocyte recruitment. IL-1B activity is normally restrained by the natural antagonist IL-1Ra and the decoy receptor IL-1R2. Dysregulated IL-1B signaling is central to autoinflammatory syndromes and contributes to many chronic inflammatory and metabolic conditions, making it a key node in innate-immune research.
