Background
The ICH5453 Crefmirlimab Biosimilar (Research Grade) is an unconjugated, non-therapeutic research analog of crefmirlimab, built around the human CD8A target (UniProt P01732). Crefmirlimab (crefmirlimab berdoxam) is best known as an anti-CD8 minibody developed as a PET immuno-imaging agent to non-invasively map CD8-positive cytotoxic T cells in tumors and lymphoid tissue, typically after radiolabeling with zirconium-89. Consistent with that biology, this product is presented in a minibody-style format (an scFv fused to a CH3 dimerization domain, indicated here as scFv-kappa-heavy with a CH3), rather than a full IgG. Supplied for research use only (RUO), it is intended as a functional and in-vitro reagent for characterizing CD8A binding rather than as a clinical imaging or therapeutic drug. As with ichor.bio in-vivo research antibodies, it is offered at low endotoxin (research grade, less than 1 EU/mg) and in bulk milligram-to-gram quantities to support method development, assay standardization, binding studies, and controls. It is not a clinical drug and carries no therapeutic claims; sequence and format should be confirmed against the buyer's intended application. CD8A (P01732) encodes the alpha chain of the CD8 co-receptor, a defining surface marker of cytotoxic T lymphocytes and a subset of dendritic and NK cells. CD8 is typically expressed as an alpha/alpha homodimer or, more commonly on conventional cytotoxic T cells, as an alpha/beta heterodimer. It binds the alpha-3 domain of MHC class I molecules, stabilizing the interaction between the T-cell receptor and peptide-MHC-I complexes and recruiting the kinase Lck to the intracellular signaling machinery, thereby lowering the threshold for T-cell activation. Because CD8 tightly marks cytotoxic effector T cells, it is a central readout in tumor immunology and immuno-oncology: CD8+ T-cell density and distribution are widely used indicators of anti-tumor immune activity and response to checkpoint therapy. This makes CD8A an attractive target for imaging and for reagents that quantify or track cytotoxic T-cell populations.
