Clone 15.2 is a mouse monoclonal antibody (isotype IgG1) recognising ICAM-1 (CD54), historically characterised against the human antigen; the listed Human/Mouse/Rat reactivity should be confirmed for the user's species before functional work. As a purified mouse IgG1, it is well suited to adhesion- and ligand-blocking studies in vitro and in vivo rather than to depletion, since IgG1 is a weak effector isotype in mouse systems. The antibody has documented utility across IHC on FFPE tissue, flow cytometry, immunoprecipitation and Western blot, supporting both target validation and functional readouts. This in-vivo grade preparation is manufactured to low endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg) and supplied in bulk milligram-to-gram quantities, making it appropriate for repeat-dosing animal studies where endotoxin contamination would otherwise confound inflammatory and leukocyte-trafficking endpoints. ICAM-1 (intercellular adhesion molecule 1, CD54) is a type I transmembrane glycoprotein of the immunoglobulin superfamily, comprising five extracellular Ig-like domains. It is expressed at low levels on endothelium, epithelium and leukocytes and is strongly upregulated by inflammatory cytokines such as TNF, IL-1 and IFN-gamma. Its principal ligands are the beta-2 integrins LFA-1 (CD11a/CD18) and Mac-1 (CD11b/CD18) on leukocytes. Through these interactions ICAM-1 mediates firm adhesion, arrest and transendothelial migration of leukocytes during inflammation, and contributes to formation of the immunological synapse between antigen-presenting cells and T cells. ICAM-1 also serves as a receptor for rhinovirus and for Plasmodium falciparum-infected erythrocytes. Because of its central role in leukocyte trafficking and immune activation, ICAM-1 is a widely studied target in inflammation, autoimmunity, ischaemia-reperfusion injury and tumour biology.
