Anti-CD8 beta In Vivo Antibody – Low Endotoxin (53-5.8) recognizes an epitope on mouse CD8b.2 and antibody clone 53-5.8 is specific for Ly-3.2 and has low reactivity with Ly-3.1
This monoclonal antibody was purified using multi-step affinity chromatography methods such as Protein A or G depending on the species and isotype.

CD8b is also known as Lyt-3.2 or Ly-3.2. It is a member of the Ig superfamily expressed as a heterodimer with the CD8A chain on a subset of MHC class I-restricted T cells and most thymocytes.

Western Blot, Functional Assays, Flow Cytometry, Immunocytochemistry, IHC (Frozen), Immunoprecipitation
Each investigator should determine their own optimal working dilution for specific applications.

Sample: Frozen sections of tumor tissues from tumor bearing C57BL / 6 mice (inoculated with LLC cells)
1. Tumors were dissected, fixed in 4% paraformaldehyde, and dehydrated in 30% sucrose;
2. Frozen tumor sections were prepared at 25 ℃ and rinsed in PBS;
3. Blocking buffer: PBS containing 0.3% Triton + 5% goat serum; Sections were blocked for 1h;
4. Primary antibodies: Diluted in blocking buffer; incubated overnight at 4℃. Final concentration of ichorbio CD8 beta antibody clone 53-5.8 (low) 3.5μg/ml, Final concentration (high) 35μg/ml. Positive signals were detected at both high and low concentrations
5. Washed by PBST; Secondary antibodies: incubated at 4℃ for 6h; DAPI: 2h

Alexa Fluor 647-AffiniPure Goat Anti-Rat IgG (H+L) (min X Hu,Bov,Hrs Sr Prot) antibody – Jackson Immunoresearch Labs Cat# 112-605-062 – Conc. 7.5μg/ml.
6. Washed by PBST at least 6 times；
7. Add fluorescence decay resistant medium, seal slice；
8. Detected by the laser scanning confocal microscope.
